Clone 1C9 is a ZooMAb^® Rabbit recombinant monoclonal antibody that specifically detects CD42b/GP1BA. It targets an epitope within 15 amino acids from the extracellular domain within the C-terminal half.

Quality Control Testing

Evaluated by Western Blotting in Human platelet cell lysates.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected CD42b/GP1BA in Human platelet cell lysate.

Tested Applications

Affinity Binding Assay: A representative lot of this antibody bound CD42b/GP1BA peptide with a KD of 1.9 x 10-6 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected CD42b/GP1BA in TF-1 cells.

Platelet glycoprotein Ib alpha chain (UniProt: P07359; also known as GP-Ib alpha, GPIb alpha, GPIbA Glycoprotein Ib alpha, Antigen CD42b-alpha, CD42b) is encoded by the GP1BA gene (Gene ID: 2811) in human. CD42b is a single-pass type I membrane glycoprotein that is synthesized with a signal peptide (aa 1-16), which is subsequently cleaved off to produce the mature protein that contains an extracellular domain (aa 17-531), a transmembrane domain (aa 532-552), and a cytoplasmic domain (aa 553-652). Following its synthesis, it may also undergo proteolytic cleavage by ADAM17 to produce Glycocalicin (aa 17-506) that contains the binding sites for von Willebrand factor (vWF) and thrombin. CD42b is expressed by platelets, and it participates in the formation of platelet plugs by binding to the A1 domain of vWF that is already bound to sub-endothelium. A transient interaction of CD42b and vWF is shown to reduce platelet velocity at sites of vascular damage and play a role in haemostasis and thrombosis. The extracellular domain of CD42b contains multiple LRR domains, some of which are involved in regulating shear-dependent adhesion to vWF. Mutations in GP1BA gene have been linked to multiple bleeding disorders, including the pseudo-von Willebrand disease (VWDP) and Bernard-Soulier syndrome (BSS) that cause prolonged bleeding time. This ZooMAb^® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
