General description

AP180 binds to clathrin triskelia with a stoichiometry of one per clathrin triskelion (trimer of heavy chains) and thereby induces assembly of clathrin into polyhedral cages. Monoclonal antibodies which react specifically with coated vesicle proteins are useful for studies on the intracellular distribution and structural relationship of clathrin-binding proteins. Mouse monoclonal anti-assembly protein 180 antibody binds to human AP180.
The antibody reacts in immunoblotting of bovine and rat brain preparations with the AP180 polypeptide (180 kDa) and with the proteolytic derivatives of AP180 (approximately 170 kDa). The antibody reacts with neuronal tissues and cultured cell lines. The antibody may be used in immunoaffinity purification. For immunocytochemistry applications, fixation with acetone or methanol is recommended.

Immunogen

membrane protein fraction from bovine brain-coated vesicles.

Application

Mouse monoclonal anti-assembly protein 180 antibody can be used for immunoblot and immunofluorescence studies. The antibody can also be used for immunocytochemistry and immunoprecipitation studies.

Physical form

The product is provided as ascites fluid with 0.1% sodium azide as a preservative.

The clathrin-assembly protein AP180 is known to promote the assembly of clathrin-coated vesicles in the neuron. However, it is unknown whether the expression of AP180 is influenced by neuronal activity. In this study, we report that chronic depolarization results in

A clathrin assembly protein (AP180) has been purified and characterized from coated vesicles of bovine brain. This protein has hitherto escaped detection because in SDS-gel electrophoresis it is obscured by the 180 kd heavy chain of clathrin. Despite the similarity

The overproduction and extracellular buildup of amyloid-beta peptide (Abeta) is a critical step in the etiology of Alzheimer's disease. Recent data suggest that intracellular trafficking is of central importance in the production of Abeta. Here we use a neuronal cell

Clathrin-coated vesicles are known to play diverse and pivotal roles in cells. The proper formation of clathrin-coated vesicles is dependent on, and highly regulated by, a large number of clathrin assembly proteins. These assembly proteins likely determine the functional specificity

Newly formed synapses undergo maturation during ontogenetic development via mechanisms that remain poorly understood. We show that maturation of the presynaptic endocytotic machinery in CNS neurons requires substitution of the adaptor protein 3 (AP-3) with AP-2 at the presynaptic plasma
