General description

Bcl-10 or CIPER is an N-terminal CARD (Caspase Recruitment Domain) containing protein mapped on to chromosome 1 at 1p22 and is localized in perinuclear region of cytoplasm and in membrane raft along with DPP4.

Immunogen

synthetic peptide corresponding to the human Bcl-10 sequence (amino acids 216-233), with N-terminal lysine added, conjugated to KLH.

Application

Polyclonal anti-Bcl-10 (AQ-18) antibody produced in rabbit is suitable for immunoprecipitation at a concentration of 5-10μg by using lysate from 4-6x105 human THP-1/Raji cells and for immunohistochemistry at 1:250 working dilution using formalin-fixed, paraffin-embedded heat-retrieved, sections of rat spleen. The product is also suitable for western blot analysis at a working dilution of 1:2,500 using whole extract of human THP-1/Raji cells and at 1:500 using whole extract of mouse NIH3T3 fibroblasts. Further, anti-Bcl-10 (AQ-18) antibody has shown suitability for microarray analysis.

Biochem/physiol Actions

Bcl-10 is a cellular homolog of the equine herpesvirus-2 protein E-10 (vCLAP). It has been implicated in the regulation of apoptosis by interacting with caspase 9, enhancing procaspase 9 processing and triggering its activation when overexpressed in the cell. Deregulation of Bcl-10 expression leads to cellular oncogenesis.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
