General description

Cdk1 is a regulatory protein that stimulates mitosis in eukaryotic cells. Phosphorylation of Thr14 and Tyr15 residues in Cdk1 causes its inactivation, which eventually results in the exit of cells from the mitotic cycle. Cdk1 may also be in involved in protein SUMOylation during cell cycle regulation . Anti-phospho-Cdk1 (p(Thr14)/p(Tyr15)) antibody is specific for rat, human and mouse Cdk1 (p(Thr14)/p(Tyr15)) and other cyclin-dependent kinases that are phosphorylated on these amino acid residues. The antibody does not bind with Cdk6 or Cdk4.

Immunogen

synthetic phosphopeptide derived from the region of the human cdk (cyclin-dependent kinases) that contains threonine 14 and tyrosine 15.

Application

Anti-phospho-Cdk1 (p(Thr14)/p(Tyr15)) antibody is suitable for use in western blot (1:1,000), immunoblot (35kDa) and immunoprecipitation (1:100).

Physical form

Solution in Dulbecco′s phosphate buffered saline (without Mg2+ and Ca2+), containing 50% glycerol with 1.0 mg/mL bovine serum albumin and 0.05% sodium azide.

Mitotic progression in eukaryotic cells depends upon the activation of cyclin-dependent kinase 1 (CDK1), followed by its inactivation through the anaphase-promoting complex (APC)/cyclosome-mediated degradation of M-phase cyclins. Previous work revealed that expression of a constitutively active CDK1 (CDK1AF) in HeLa

Increasing evidence has pointed to an important role of SUMOylation in cell cycle regulation, especially for M phase. In the current studies, we have obtained evidence through in vitro studies that the master M phase regulator CDK1/cyclin B kinase phosphorylates

The kinase activity of human p34cdc2 is negatively regulated by phosphorylation at Thr-14 and Tyr-15. These residues lie within the putative nucleotide binding domain of p34cdc2. It has been proposed that phosphorylation within this motif ablates the binding of ATP
