Application
Anti-Caspase 8 (GD-13) is suitable for microarray. It is also suitable for western blot at a concentration of 1:1,000 using human Burkitt lymphoma Raji cell extract.
Biochem/physiol Actions
Caspase 8 cleaves and activates CASP3, CASP4, CASP6, CASP7, CASP9 and CASP10. May participate in the GZMB apoptotic pathways. It also cleaves ADPRT. Caspase 8 hydrolyzes the small-molecule substrate, Ac-Asp-Glu-Val-Asp-|-AMC. It is also a target for cowpox virus CRMA death inhibitory protein. Anti-Caspase 8 (GD-13) recognizes human caspase 8a (55 kDa) and caspase 8b (54kDa) by immunoblotting. Staining of caspase 8 in immunoblotting is specifically inhibited with caspase 8 immunizing peptide (human, amino acids 362-374). Caspases play a crucial role in apoptosis in almost every cell type.
The sequence used as the immunogen corresponds to the C-terminal region of the enzyme p18 subunit and is identical in other human caspase 8 isoforms (MACHα2, MACHα3, MACHβ) and has limited homology (approx. 50%) to mouse caspase 8. By immunoblotting, the antibody reacts with caspase 8a and caspase 8b.
Physical form
Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
