General description
ColorWheel® technology is a novel and proprietary method of creating your own antibody and dye combinations for use in flow cytometry. By incubating any ColorWheel® antibody with any ColorWheel® dye, researchers can quickly and simply produce primary conjugated antibodies for use in single color or multicolor flow cytometry analysis. Each ColorWheel® antibody and dye is lyophilized for long long-term storage, allowing for simplicity and flexibility without compromise. ColorWheel® technology requires both ColorWheel® antibody and ColorWheel® dye for Flow Cytometry application.
Early activation antigen CD69 (UniProt: Q07108; Activation inducer molecule, AIM, BL-AC/P26, C-type lectin domain family 2 member C, EA1, Early T-cell activation antigen p60, GP32/28, Leukocyte surface antigen Leu-23, MLR-3, CD69) is encoded by the CD69 (also known as CLEC2C) gene (Gene ID: 969) in human. CD69 is a single-pass type II membrane disulfide-linked homodimeric protein with a cytoplasmic domain (aa 1-40), a transmembrane domain (aa 41-61), and an extracellular domain (aa 62-199). It is a C-type lectin and a member of the natural killer (NK) receptor family. It is an early activation marker that is expressed in hematopoietic stem cells, activated T-cells, B-cells, natural killer cells, neutrophils, eosinophils, epidermal Langerhans cells, and platelets. It is induced by antigens, mitogens, or activators of PKC on the surface of T and B-lymphocytes and by interaction of IL-2 with the p75 IL-2R on the surface of NK cells. CD69 is involved in immune cell homeostasis and regulates T cell-mediated immune response through the control of Th17 cell differentiation. Expression of CD69 by activated T lymphocytes is reported to trigger an anti-inflammatory mechanism mediated by Galectin-1, which regulates the immune response and prevents pathogenic Th17 responses. Staining of one million human peripheral blood mononuclear cells (PBMC) stimulated with Ionomycin/PMA (1 μg/mL, 4 h) was performed using 5 μL of a 1:1 mixture of Cat. No. CWA-1037, Anti-Human CD69 (FN50) ColorWheel® Dye-Ready mAb and Cat. No. CWD-PE ColorWheel® Antibody-Ready Phycoerythrin (PE) Dye or an equivalent amount of PE-conjugated Mouse IgG1 isotype control.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user

Biochem/physiol Actions
Clone FN50 is a mouse monoclonal antibody that detects human CD69.
Physical form
Lyophilized from PBS containing D-Mannitol and Sucrose, normal appearance is a dried pellet. Reconstituted antibody solution is stable and functional as assessed by functional testing. Contains no biocide or preservatives, such as azide.
Preparation Note
1.0 mg/mL after reconstitution at 25 μL in PBS. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C. Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial. Reconstitute each vial by adding 25 μL of PBS. Protect from light.
