Anti-Mouse MHC II (M5/114.15.2) ColorWheel^® Dye-Ready mAb

ColorWheel^® technology is a novel and proprietary method of creating your own antibody and dye combinations for use in flow cytometry. By incubating any ColorWheel^® antibody with any ColorWheel^® dye, researchers can quickly and simply produce primary conjugated antibodies for use in single color or multicolor flow cytometry analysis. Each ColorWheel^® antibody and dye is lyophilized for long-term storage, allowing for simplicity and flexibility without compromise. ColorWheel^® technology requires both ColorWheel^® antibody and ColorWheel^® dye for Flow Cytometry application.

H-2 class II histocompatibility antigen, A-K alpha chain/H-2 class II histocompatibility antigen, A beta chain/H-2 class II histocompatibility antigen gamma chain (UniProt: P01910/ P14483/P04441) is encoded by the H2-Aa/H2-Ab1/Cd74 gene (Gene ID: 14960/14961/16149) in murine species. MHC Class II molecules are a class of major histocompatibility complex (MHC) molecules found on antigen-presenting cells such as dendritic cells, mononuclear phagocytes, some endothelial cells, thymic epithelial cells, and B cells. Their major function is to present processed antigens, derived primarily from exogenous sources, to CD4+ T-lymphocytes. MHC-II is a heterodimeric transmembrane glycoprotein comprising an subunit associated non-covalently with a β subunit. In mice two isoforms of MHC Class II have been described and are known as I-A and I-E and they can form A /Aβ and E /Eβ heterodimers. The newly synthesized and β chains assemble into heterodimers in the endoplasmic reticulum, where they are bound by trimers of invariant chain (g), which serves as a dedicated chaperone. They form nonamers of three a/b/g heterotrimers that traffic into late-stage endosomal compartment where the invariant chain is degraded to form CLIP (class II invariant chain-associated peptide). CLIP temporarily occupies the peptide binding groove of the MHC class II molecules. A catalytic protein, HLA-DM, then exchanges CLIP for peptides that bind MHC class II with high-affinity. The peptide/MHC class II complex can then traffic to the cell surface for interaction with CD4+ T cells through the T-cell receptor, which serves as the first signal to the lymphocytes to elicit an antigen-specific adaptive response. Clone M5/114.15.2 reacts with mouse MHC Class II alloantigens I-Ab, I-A d, I-Aq, I-Ed, and I-Ek and can cross-react with mouse cells of H-2p and H-2r haplotype. (Ref.: Wosen, JE., et al. (2018). Flow Cytometry Analysis: Staining of one million mouse splenocytes was performed using 5 μL of a 1:1 mixture of Cat. No. CWA-1081-25UL, Anti-Mouse MHC II (M5/114.15.2) ColorWheel^® Dye-Ready mAb and Cat. No. CWD-PE ColorWheel^® Antibody-Ready Phycoerythrin (PE) Dye or an equivalent amount of PE-conjugated Rat IgG2b isotype control.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user

Clone M5/114.15.2 is a rat monoclonal antibody that detects MHC Class II; Major Histocompatibility Class II (MHC II).

Lyophilized from PBS containing D-Mannitol and Sucrose, normal appearance is a dried pellet. Reconstituted antibody solution is stable and functional as assessed by functional testing. Contains no biocide or preservatives, such as azide.

1.0 mg/mL after reconstitution at 25 μL in PBS. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Recommend storage of lyophilized product at 2-8°C. Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial. Reconstitute each vial by adding 25 μL PBS. Protect from light.
