General description
ColorWheelColorWheel® technology is a novel and proprietary method of creating your own antibody and dye combinations for use in flow cytometry. By incubating any ColorWheelColorWheel® antibody with any ColorWheelColorWheel® dye, researchers can quickly and simply produce primary conjugated antibodies for use in single color or multicolor flow cytometry analysis. Each ColorWheelColorWheel® antibody and dye is lyophilized for long-term storage, allowing for simplicity and flexibility without compromise. ColorWheelColorWheel® technology requires both ColorWheelColorWheel® antibody and ColorWheelColorWheel® dye for Flow Cytometry application.
Integrin alpha-L (UniProt: P24063; also known as CD11 antigen-like family member A, Leukocyte adhesion glycoprotein LFA-1 alpha chain, LFA-1A, Leukocyte function-associated molecule 1 alpha chain, Lymphocyte antigen 15, Ly-15, CD11a) is encoded by the Itgal (also known as Lfa-1, Ly-15) gene (Gene ID: 16408) in murine species. Integrins are part of a family of heterodimeric transmembrane glycoproteins that recognize a variety of ligands and control numerous physiologic functions in cells. CD11a is a single-pass type I membrane glycoprotein protein that is synthesized with a signal peptide (aa 1-23), which is subsequently cleaved off in the mature form that contains a large extracellular domain (aa 24-1087), a transmembrane domain (aa 1088-1108), and a cytoplasmic domain 1109-1163). CD11a noncovalently associates with b2 integrin to forms a heterodimer to serve as a receptor for ICAM1, ICAM2, ICAM3 and ICAM4. It is expressed on B and T lymphocytes, monocytes, macrophages, neutrophils, basophils, and eosinophils. It is also involved in generation of common lymphoid progenitor cells in bone marrow. In resting T-cells, about 40% of CD11a is constitutively phosphorylated and phosphorylation causes conformational changes needed for ligand binding and is considered to be essential for activation by some physiological agents. CD11a -/- mice exhibit decreased thymic cellularity and impaired early T cell development. CD11a is shown to be critical for generation of common lymphoid progenitors (CLPs) and lymphoid-primed multipotent progenitors (LMPPs). (Ref.: Bose, TO., et al. (2014). J. Biochem/physiol Actions
Clone M17/4 is a rat monoclonal antibody that detects murine CD11a (Integrin aL). It targets an epitope within the extracellular domain.
Physical form
Lyophilized from PBS containing D-Mannitol and Sucrose, normal appearance is a dried pellet. Reconstituted antibody solution is stable and functional as assessed by functional testing. Contains no biocide or preservatives, such as azide.
Preparation Note
1.0 mg/mL after reconstitution at 25 μL in PBS. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C. Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial. Reconstitute each vial by adding 25 μL PBS. Protect from light.
