ColorWheel® technology is a novel and proprietary method of creating your own antibody and dye combinations for use in flow cytometry. By incubating any ColorWheel® antibody with any ColorWheel® dye, researchers can quickly and simply produce primary conjugated antibodies for use in single color or multicolor flow cytometry analysis. Each ColorWheel® antibody and dye is lyophilized for long-term storage, allowing for simplicity and flexibility without compromise. ColorWheel® technology requires both ColorWheel® antibody and ColorWheel® dye for Flow Cytometry application.
Natural killer cells antigen CD94 (UniProt: Q13241; also known as KP43, Killer cell lectin-like receptor subfamily D member 1, NK cell receptor, CD94) is encoded by the CD94 (also known as KLRD1) gene (Gene ID: 3824) in human. CD94 is a single-pass type II membrane protein that contains a cytoplasmic domain (aa 1-10), a transmembrane domain (aa 11-31), and an extracellular domain (aa 32-179). It also contains a C-type lectin domain (aa 68-175). CD94 expression is observed in subsets of NK cells, memory/effector CD8+ , β T cells, and melanoma-specific cytotoxic T cells. It is also expressed in terminally differentiated cytotoxic , T cells. It serves as an immune inhibitory receptor that is involved in self-non-self-discrimination. It serves primarily as a ligand binding subunit as it lacks the capacity to signal. In complex with KLRC1 (Cd159a) or KLRC2 (CD159c) on cytotoxic and regulatory lymphocyte subsets, it recognizes non-classical major histocompatibility (MHC) class Ib molecule HLA-E that is loaded with self-peptides derived from the signal sequence of classical MHC class Ia and non-classical MHC class Ib molecules. It enables cytotoxic cells to monitor the expression of MHC class I molecules in healthy cells and to tolerate self. CD94 is also reported to dominantly counteract T cell receptor signaling on a subset of memory/effector CD8+ T cells as part of an antigen-driven response to avoid autoimmunity. Flow Cytometry Analysis: Staining of one million human peripheral blood mononuclear cells (PBMC), lymphocytes was performed using FITC conjugated anti-human CD56 and 5 µL of a 1:1 mixture of Cat. No. CWA-1165, Anti-Human CD94/KLRD1 (HP-3D9) ColorWheel® Dye-Ready mAb and Cat. No. CWDS-PE ColorWheel® Antibody-Ready Phycoerythrin (PE) Dye or an equivalent amount of Cat. No. CWA-1013 Mouse IgG1 isotype control and Cat. No. CWDS-PE ColorWheel® Antibody-Ready Phycoerythrin (PE) Dye. CD94 expression was observed mainly in CD56 positive population.
Tested Applications

No. CWA-1013 Mouse IgG1 isotype control and Cat. No. CWDS-APC ColorWheel® Antibody-Ready Allophycocyanin (APC) Dye. CD94 expression was observed mainly in CD56 positive population.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Lyophilized from PBS containing D-Mannitol and Sucrose, normal appearance is a dried pellet. Reconstituted antibody solution is stable and functional as assessed by functional testing. Contains no biocide or preservatives, such as azide.
制备说明
1.0 mg/mL after reconstitution at 25 µL in PBS. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C. Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial. Reconstitute each vial by adding 25 µL PBS. Protect from light.
