ColorWheel^® technology is a novel and proprietary method of creating your own antibody and dye combinations for use in flow cytometry. By incubating any ColorWheel^® antibody with any ColorWheel^® dye, researchers can quickly and simply produce primary conjugated antibodies for use in single color or multicolor flow cytometry analysis. Each ColorWheel^® antibody and dye is lyophilized for long-term storage, allowing for simplicity and flexibility without compromise. ColorWheel^® technology requires both ColorWheel^® antibody and ColorWheel^® dye for Flow Cytometry application.

CD70 antigen (UniProt: P32970; also known as CD27 ligand, CD27-L, Tumor necrosis factor ligand superfamily member 7, CD70) is encoded by the CD70 (also known as CD27L, CD27LG, TNFSF7) gene (Gene ID: 970) in human. CD70 is a single-pass type II, homotrimeric, membrane glycoprotein that contains a cytoplasmic domain (aa 1-17), a transmembrane domain (aa 18-37), and an extracellular domain (aa 39-193). It is a member of the TNF family whose expression is observed in antigen-presenting cells and is strongly but transiently up-regulated during lymphocyte activation. It is expressed on dendritic cells during their maturation and plays a critical role in immunogenicity and in priming T cells. It serves as a ligand for CD27 that is expressed on T cells and NK cells. CD70-CD27 pathway plays an important role in the generation and maintenance of T cell immunity, in particular during antiviral responses. The CD70-CD27 pathway promotes effector CD8+ T cells responses and influences polarization of CD4+ T cells. Upon binding to CD27, it induces the proliferation of co-stimulated T-cells and enhances the generation of cytolytic T-cells. CD70 also has a regulatory role in early cytokine responses to pathogens through the maintenance of Treg cell survival and function. CD70 and CD27 are aberrantly expressed in many hematological and solid malignancies and the dysregulation of the CD70-CD27 axis within the tumor and its microenvironment is associated with tumor progression and immunosuppression. Mutations in CD70 gene are known to cause lymphoproliferative syndrome 3, an autosomal recessive disorder that is characterized by increased susceptibility to Epstein-Barr virus infection in B cells, abnormal B-cell proliferation, and increased susceptibility to B-cell malignancies. Flow Cytometry Analysis: Staining of one million HuT 78 cells was performed using 5 µL of a 1:1 mixture of Cat. No. CWA-1174, Anti-Human CD70 (Ki-24) ColorWheel^® Dye-Ready mAb and Cat. No. CWDS-PE ColorWheel^® Antibody-Ready Phycoerythrin (PE) Dye or an equivalent amount of PE-conjugated Mouse IgG3 isotype control.

Tested Applications

Flow Cytometry Analysis: Staining of one million HuT 78 cells was performed using 5 µL of a 1:1 mixture of Cat. No. CWA-1174, Anti-Human CD70 (Ki-24) ColorWheel^® Dye-Ready mAb and Cat. No. CWDS-FITC ColorWheel^® Antibody-Ready FITC Dye or an equivalent amount of FITC conjugated Mouse IgG3 isotype control.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Clone Ki-24 is a mouse monoclonal antibody that detects Human CD70. It targets an epitope within the extracellular domain.

Lyophilized from PBS containing D-Mannitol and Sucrose, normal appearance is a dried pellet. Reconstituted antibody solution is stable and functional as assessed by functional testing. Contains no biocide or preservatives, such as azide.

1.0 mg/mL after reconstitution at 25 µL in PBS. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Recommend storage of lyophilized product at 2-8°C. Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial. Reconstitute each vial by adding 25 µL PBS. Protect from light.
