General description

17β-estradiol (E2) is a sex hormone that is a key regulator of growth, differentiation and function of reproductive tissues, skeletal and cardiovascular systems. The effect of 17β-estradiol is mediated by two intracellular receptors, ERα and ERβ. Although the two receptors interact with similar ligands, have identical DNA-binding domains and have redundant roles in estrogen signalling, they show distinct expression pattern. On binding to the receptor, the estrogen-ER complex enters nucleus and regulates the transcription of specific genes. The biological effects of estradiol may be mediated by the classic binding of estrogen and ER, by upstream activation of the pathway by growth factors. Some pathways that activate the estrogen-ER pathway are those mediated by AP-1, IGF-1, EGF, MAPK and PI3K/Akt.

Application

Anti-17β-Estradiol antibody may be used for RIA. The antibody has also been used for ELISA.

Preparation Note

Reconstitute with 5 ml buffer: 0.05 M Tris-HCl, pH 8.0, 0.1 M sodium chloride, 0.1% gelatin and 15 mM sodium azide. Store this stock at −20 °C. Working dilution (1:10) to be discarded if not used within 12 hours.
