General description
EphA7 (ephrin receptor A7) is a receptor tyrosine kinase (RTK), which belongs to a 16 member family of ephrin receptors. This family is divided into two classes A and B. EphA5 belongs to EphA group, which contains 10 members. The ligands to this group of receptors are glycosylphosphatidylinositol (GPI)-anchored named ephrin-A1-A6. The members of this family have an ephrin-binding domain in its N-terminal globular domain, succeeded by a linker region rich in cysteine and two fibronectin type III repeats in the exoplasmic domain. Its cytoplasmic domain consists of a juxtamembrane region, a conserved tyrosine kinase domain, an α-helical C-terminal domain, and a PDZ domain-binding domian.
Biochem/physiol Actions
EphA7 (ephrin receptor A7) promotes normal synaptic functioning, and development of cortical dendrites and maturation of the spine, and acts as a guidance receptor during development and patterning of cortex. During initial stages of brain development, this receptor modulates cell apoptosis. It forms a multi-protein complex with tumor necrosis factor receptor 1 (TNFR1) and EphA5 to induce caspase-mediated cell death. In mice, it is induced by Dlx5 and Msx2, which are homeodomain-containing transcription factors. It therefore, regulates the morphogenesis of cranial neural tube, by controlling the cell adhesion. In mice, it also regulates the contouring of corticothalamic (CT) mapping. It plays the role of tumor-suppression in follicular lymphoma (FL), and thus has potential as a therapeutic target.
