一般描述
谷蛋白由醇溶蛋白和谷蛋白两个醇溶蛋白家族组成。醇溶蛋白是一种疏水性很强的糖蛋白,溶解性极差。醇溶蛋白可进一步细分为 α-醇溶蛋白类,γ-醇溶蛋白和 ω-醇溶蛋白(基于其在低 pH 下电泳时的迁移率顺序。
应用
小麦来源麦醇溶蛋白用于:
- 胃蛋白酶-胰蛋白酶消化,研究体外麦醇溶蛋白挑战对无麸质饮食(GFD)乳糜泻(CD)患者活检组织的影响
- 制备麦醇溶蛋白,用于抗麦醇溶蛋白抗体测定

生化/生理作用
麦醇溶蛋白包含富含谷氨酰胺和脯氨酸的单体蛋白质家族,其占小麦籽粒总蛋白质含量的 40-50%。 醇溶性麦醇溶蛋白与不溶于醇的谷蛋白聚集形成谷蛋白,这是小麦面团的结构和性质的原因。 麦醇溶蛋白还参与乳糜泻的发病机制,乳糜泻是一种自身免疫性疾病,其中人白细胞抗原 DQ2 和 DQ8 将胶质肽递呈至 T 细胞亚群,诱导促炎细胞因子的释放,进而诱导肠黏膜基质的降解和细胞死亡。B 细胞也被刺激产生谷蛋白特异性和转谷氨酰胺酶 2 (TG2) 特异性抗体,后者可能是由麦醇溶蛋白肽与 TG2 共价结合的呈递所致。
