General description
Intermediate filaments (IFs) have a diameter of 8-10nm and differ from microtubules and microfilaments significantly. Neurofilaments (NFs) are a type of IFs found mainly in cells or tissues of neuronal origin. NFs are composed of three major proteins, NF-L, NF-M and NF-H, having a molar mass of 68kDa, 160kDa, and 200kDa, respectively.
Monoclonal Anti-O-Glycosylated Neurofilament-M (mouse IgG2a isotype) is derived from the hybridoma NL6 produced by the fusion of mouse myeloma cells and splenocytes from BALB/c mice immunized with a protein fraction enriched for components of the human neuronal cytoskeleton. Neurofilament medium (NEFM) gene encodes neurofilament medium polypeptide which constitutes to cytoskeleton of mature neurons. The gene in human chromosome is localized on 8p21.
Immunogen
protein fraction enriched for components of the human neuronal cytoskeleton from cell lines NT2-N and SK-N-BE(2). The epitope is localized in the carboxy-terminal tail domain of NF-M.

Biochem/physiol Actions
Neurofilament medium (NEFM) is essential for radial growth of axon which includes the diameter of the axon as well as the myelination. The C-terminus of NFEM gene has 7 repeats of KSP (lysine-serine-proline) domain and phosphorylation of which is crucial for regulating the radial growth of axon.
The NF proteins are synthesized in the neuronal perikarya before assemby to form the filaments. These filaments are then transported within the axons towards the synaptic terminals. The function of these NF polypeptides is regulated by phosphorylation, which influences their interaction with cytoplasmic organelles. The head region of NF-L and NF-M contain sites modified by O-linked GlcNAc. Down-regulation of NF-M O-GlcNAcylation and increased phosphorylation of NF-M is observed in Alzheimer disease. This antibody recognizes specifically an O-glycosylated epitope in the projection domain of NF-M. In a rat model for amyotrophic lateral sclerosis (ALS), this specific modification is strongly decreased compared with wild-type animals.
