Biochem/physiol Actions
Pre-mRNA-splicing factor SPF27 is a protein encoded by the BCAS2 gene in humans and is mapped to chromosome 1p13.3-21 region. It codes for a nuclear protein. BCAS2 is a novel gene up-regulated by amplification in human breast cancer cell lines. It acts as a transcriptional coactivator of estrogen receptor. It directly interacts with p53 to reduce p53 transcriptional activity and consistently decreases p53 protein in the absence of DNA damage. BCAS2 is a small nuclear protein (26kDa) and a novel negative regulator of p53. It is a potential molecular target for cancer therapy.

BCAS2 is a novel gene isolated from breast cancer cell line by differential display technique. Previously we reported that BCAS2 gene is localized on chromosome 1p13.3-21 and is up-regulated by gene amplification in breast cancer cell lines MCF-7 and BT-20.

Breast cancer amplified sequence 2 (BCAS2) was reported previously as a transcriptional coactivator of estrogen receptor. Here, we report that BCAS2 directly interacts with p53 to reduce p53 transcriptional activity by mildly but consistently decreasing p53 protein in the absence

Screening for differentially expressed genes in cancer cell lines by the RNA differential display (DD) technique identified a novel mRNA that encodes a 26-kDa protein and is up-regulated in MCF-7 and BT-20 breast cancer cells. The predicted amino acid sequence
