生化/生理作用

Hhip (hedgehog-interacting protein) from mouse is a membrane-associated glycoprotein and an antagonist of hedgehog (Hh) signalling. Mice embryos lacking HIP1 have lung defects and die after birth. Hip interacts with mammalian hedgehog family proteins, namely Shh (sonic hedgehog), Ihh (indian hedgehog) and Dhh (desert hedgehog). It is transcriptionally regulated by Hh signalling. Hh proteins are crucial for controlling cell proliferation and cell fate.

特点和优势

Based on N-terminal sequencing, this preparation contains a mixture of at least two peptides that have Lys 24 and Asp 52, respectively, at the N-terminus. The two peptides have calculated molecular masses of ~76 kDa and 73 kDa, respectively. Due to glycosylation and C-terminal proteolytic processing, recombinant mouse Hip migrates with apparent molecular masses of 80-90 kDa and 58-64 kDa, respectively, in SDS-PAGE under reducing conditions.

外形

Lyophilized from a 0.2 μm filtered solution in phosphate buffered saline containing 2.5 mg bovine serum albumin.

分析说明

The biological activity is measured by its ability to inhibit sonic hedgehog (Shh) induction of alkaline phosphatase production in C3H10T1/2 fibroblasts.

The Hedgehog signalling pathway is essential for the development of diverse tissues during embryogenesis. Signalling is activated by binding of Hedgehog protein to the multipass membrane protein Patched (Ptc). We have now identified a novel component in the vertebrate signalling

Hedgehog-interacting protein (Hhip) is a novel regulatory component in the vertebrate hedgehog-signalling pathway. The murine Hhip encodes a type I TM protein that attenuates hedgehog signalling by binding all three mammalian hedgehog proteins. Here we describe the cloning and characterisation

The Hedgehog (Hh) signaling pathway regulates a variety of developmental processes, including vasculogenesis, and can also induce the expression of pro-angiogenic factors in fibroblasts postnatally. Misregulation of the Hh pathway has been implicated in a variety of different types of
