Immunogen
Ancient ubiquitous protein 1 precursor recombinant protein epitope signature tag (PrEST)
Application
Anti-AUP1 antibody produced in rabbit, a Prestige Antibody, is developed and validated by the Human Protein Atlas (HPA) project . Each antibody is tested by immunohistochemistry against hundreds of normal and disease tissues. These images can be viewed on the Human Protein Atlas (HPA) site by clicking on the Image Gallery link. The antibodies are also tested using immunofluorescence and western blotting. To view these protocols and other useful information about Prestige Antibodies and the HPA, visit sigma.com/prestige.
Applications in which this antibody has been used successfully, and the associated peer-reviewed papers, are given below.

Biochem/physiol Actions
AUP1 (ancient ubiquitous protein 1) gene is localized to human chromosome 2p13. It localizes to lipid droplets (LD) and integrates into the LD surface with both terminus on the cytosolic end. It contains a G2BR (G2 binding region) domain at the C-terminus that binds to Ube2g2 (E2 Ubiquitin Conjugase G2) and forms a complex, which is involved in the cellular ubiquitination machinery. The CUE domain of Aup1 binds to the HRD1-SEL1L complex and regulates polyubiquitylation. It is also localized to the endoplasmic reticulum (ER), where it participates in ER protein quality control by polyubiquitylation and degradation of misfolded proteins. It mediates the regulation of Rtg3, a transcription factor involved in the retrograde signaling pathway, during mitophagy, an autophagic process that degrades mitochondria.
Features and Benefits
Prestige Antibodies® are highly characterized and extensively validated antibodies with the added benefit of all available characterization data for each target being accessible via the Human Protein Atlas portal linked just below the product name at the top of this page. The uniqueness and low cross-reactivity of the Prestige Antibodies® to other proteins are due to a thorough selection of antigen regions, affinity purification, and stringent selection. Prestige antigen controls are available for every corresponding Prestige Antibody and can be found in the linkage section.
Every Prestige Antibody is tested in the following ways:
Every Prestige Antibody is tested in the following ways:
- IHC tissue array of 44 normal human tissues and 20 of the most common cancer type tissues.
- Protein array of 364 human recombinant protein fragments.

Sterol-induced binding to Insigs in the endoplasmic reticulum (ER) allows for ubiquitination of 3-hydroxy-3-methylglutaryl coenzyme A reductase, the rate-limiting enzyme in cholesterol synthesis. This ubiquitination marks reductase for recognition by the ATPase VCP/p97, which mediates extraction and delivery of reductase
