General description
SH3 domain-containing guanine exchange factor (ARHGEF26) is a Rho (a GTPase) guanine exchange factor. It is expressed in many cell lines and strongly expressed in liver tissue. ARHGEF26 is an 871-amino acid protein with an amino-terminal region, a DbI homology (DH) domain and a carboxyl-terminal SH3 domain. The gene encoding this protein is located on chromosome 3q25.2.

Biochem/physiol Actions
SH3 domain-containing guanine exchange factor (ARHGEF26) promotes epidermal growth factor receptor (EGFR) stability and epidermal growth factor (EGF)-induced extracellular signal-regulated kinase (ERK1/2) activation. It binds with growth factor receptor-bound protein 2 (Grb2) which is a downstream transducer of EGFR. In fibroblasts, ARHGEF26 induces the formation of interconnected cell ruffles with actin polymerization across dorsal surfaces. It colocalizes with filamentous actin at the sites where membrane protrusion has occurred. Overexpression of ARHGEF26 has been linked to prostate cancer progression.
