General description

Monoclonal Anti-Kaiso (mouse IgG1 isotype) is derived from the 6F hybridoma produced by the fusion of mouse myeloma cells and splenocytes from A/J mice. Kaiso is a DNA-binding protein and a member of the broad complex, tramtrak, bric a brac/ pox zinc finger (BTB/POZ) family of transcription factors with zinc finger motifs. This protein contains three carboxy-terminal zinc-finger motifs of the Kruppel-like C2H2-DNA binding type zinc fingers and a poxvirus and zinc finger (POZ) domain in its extreme N-terminus, which is highly hydrophobic. The zinc finger and BTB domain containing 33 (ZBTB33) gene is located on the human chromosome at Xq24.

Immunogen

recombinant purified mouse Kaiso protein (amino acids 1-499).

Application

Anti-Kaiso antibody, Mouse monoclonal may be used in:
  • enzyme-linked immunosorbent assay (ELISA)
  • immunoblotting
  • immunoprecipitation
  • immunocytochemistry

Biochem/physiol Actions

Kaiso is a transcriptional repressor that recognizes specifically the DNA sequence TCCTGCNA or two symmetrically methylated CpG dinucleotides. This protein binds to the cell adhesion cofactor p120 ctn which signifies its role in the p120 ctn action and signaling.
Monoclonal Anti-Kaiso reacts specifically with human, rat, mouse, canine, and chicken Kaiso (approx.105 kDa).

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, and 15 mM sodium azide.

Preparation Note

For continuous use, store at 2-8 °C for up to one month. For extended storage, freeze in working aliquots. Repeated freezing and thawing is not recommended. Storage in "frost-free" freezers is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilution samples should be discarded if not used within 12 hours.

The p120(ctn)-binding partner Kaiso is a new member of the POZ-zinc finger family of transcription factors implicated in development and cancer. To understand the role of Kaiso in gene regulation and p120(ctn)-mediated signaling and adhesion, we sought to identify Kaiso-specific

p120(ctn) is an Armadillo repeat domain protein with structural similarity to the cell adhesion cofactors beta-catenin and plakoglobin. All three proteins interact directly with the cytoplasmic domain of the transmembrane cell adhesion molecule E-cadherin; beta-catenin and plakoglobin bind a carboxy-terminal
