General description
Metastasis-associated protein (MTA2) comprises bromo-adjacent homology (BAH), Egl-27 and MTA1 homology 2 (ELM2), Swi3, Ada2, N-Cor, and TFIIIB (SANT), and an atypical GATA zinc finger domain. It is a phosphorylated protein. MTA2 gene is mapped to human chromosome 11q12.3. Human MTA2 shares approximately 80% overall homology to human MTA1 and MTA3 proteins, with the C-terminus being more divergent than the N-terminus.
Immunogen
synthetic peptide corresponding to amino acids 626-641 of human MTA2/MTA1L1, conjugated to KLH via an N-terminal added cysteine residue. The immunizing peptide is conserved in human, mouse, and rat, and is not present in other members of the family.
Application
Anti-MTA2/MTA1L1 (RT-16) antibody produced in rabbit has been used in:
- tandem immunoprecipitation
- immunoblotting
- immunofluorescence
Biochem/physiol Actions
Anti MTA2/MTA1L1 (RT-16) specifically recognizes MTA2/MTA1L1.
Metastasis-associated protein (MTA2) mediates cytoskeletal reorganization. It interacts with the histone deacetylases (HDAC) within the nuclear remodeling and deacetylation complexes namely, nucleosome remodeling and deacetylase (Mi2/NuRD). This interaction mediates transcriptional repression. Also called p53 target protein in deacetylase complex (PID), it regulates p53 function and represses p53-dependent transcriptional activation, growth arrest and apoptosis. MTA2 also favors tumor growth and migration.
Physical form
Solution in 0.01 M phophate buffered saline, pH 7.4, containing 15 mM sodium azide.
Preparation Note
For continuous use, store at 2-8 °C for up to one month. For extended storage, freeze in working aliquots. Repeated freezing and thawing is not recommended. Storage in frost-free freezers is also not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilutions should be discarded if not used within 12 hours.
