General description
嗜热菌蛋白酶是一种蛋白酶，其特异性不同于可用于序列研究的其他蛋白酶。
Application
一种含有四个钙离子的热稳定性（嗜热性）细胞外金属内肽酶。辅助因子是锌和钙。水解疏水性氨基酸残基N-末端的蛋白质键。最适pH为8.0，活性的最适温度为70℃。在pH 5-9.5范围内非常稳定。嗜热菌蛋白酶具有较低的切割特异性，因此，它能产生许多适合测序的短片段。优先切割顺序： X-切割-Y-Z，其中X = 任何氨基酸；Y = Leu，Phe，Ile，Val，Met，Ala；Z是除Pro以外的任何氨基酸。切割Leu的N末端优先于切割Phe的N末端，它们优于其他氨基酸。常用于限制性蛋白水解以绘制肽图谱以及蛋白质结构和构象变化研究。
嗜热菌蛋白酶也常用于商业合成N-（苄氧基羰基）-L-天冬氨酰-L-苯丙氨酸甲酯——人造甜味剂阿斯巴甜的前体。
已有研究显示嗜热菌蛋白酶具有前序列，可在体内充当分子内分子伴侣。 它还被用于研究氯化钠对其热稳定性和催化活性的影响。
