General description
RAD1 checkpoint DNA exonuclease/RAD1 is located on human chromosome 5p13.2. It belongs the Rad family proteins.
Immunogen
synthetic peptide corresponding to amino acids 267-282 of human RAD1, conjugated to KLH via an N-terminal added lysine residue. The immunizing peptide is present in RAD1A and RAD1B (isoforms 1 and 2).
Application
Anti-RAD1, C-terminal antibody produced in rabbit may be used in immunoblotting and immunoprecipitation.
Biochem/physiol Actions
Anti RAD1 (C-terminal) specifically recognizes RAD1.
RAD1 checkpoint DNA exonuclease/RAD1 can form a stable radioresponsive checkpoint complex commonly known as 9-1-1, which participates in cellular responses to DNA damage or inhibition of DNA synthesis. Rad1 can act as a DNA damage sensor. RAD1 exhibits terminal exonuclease activity on double-stranded DNA with a preference for 3′ ends. It can regulate specifically the expansion of minisatellite tracts.
Physical form
Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
Preparation Note
For continuous use, store at 2-8 °C for up to one month. For extended storage, freeze in working aliquots. Repeated freezing and thawing, or storage in “frost-free” freezers, is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilutions should be discarded if not used within 12 hours.
