General description
RAD9 checkpoint clamp component A (RAD9A), an important member of the 911 complex is mapped to human chromosome 11q13.
Immunogen
synthetic peptide corresponding to amino acid residues 376-391 of human RAD9, conjugated to KLH via an N-terminal added cysteine residue. The immunizing peptide corresponds to RAD9A and is not present in RAD9B.
Application
Anti-RAD9 antibody produced in rabbit may be used in western blotting.

Biochem/physiol Actions
Anti-RAD9 specifically recognizes recombinant RAD9 (50 and 60 kDa).
RAD9 checkpoint clamp component A (RAD9A) participates in human cellular responses to DNA damage. RAD9 is found to possess 3′ to 5′ exonuclease activity, which may contribute to its role in sensing and repairing DNA damage. RAD9 can be phosphorylated by ataxia-telangiectasia mutated (ATM) in response to DNA damage. It helps to stimulate S-phase checkpoint by modulating the phosphorylation of cell cycle checkpoint protein (RAD17), checkpoint kinases (Chks 1 and 2) Chk1, Chk2 and structure maintenance of chromosome 1 (Smc1) in response to replication block and DNA damage. It helps to maintain the stability of genome. In addition to its DNA repair role, it also plays a major role in radio resistance, androgen receptor signaling repression and immunoglobulin class switching. RAD9 also participates in telomere maintenance.

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
Preparation Note
For continuous use, store at 2-8 °C for up to one month. For extended storage, freeze in working aliquots. Repeated freezing and thawing, or storage in “frost-free” freezers, is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilutions should be discarded if not used within 12 hours.
