Gene Information
rat ... Magi3(245903)
General description
Monoclonal Anti- SLIPR/MAGI-3 (mouse IgG1 isotype) is derived from the SLP-32 hybridoma produced by the fusion of mouse myeloma cells and splenocytes from mice. Membrane-associated guanylate kinase, WW and PDZ domain-containing 3 (MAGI-3) protein belong to the membrane-associated guanylate kinase (MAGUK) family. This protein is made up of a guanylate-kinase-like region, six post-synaptic density-95/discs large/zonula occludens-1 (PDZ), and two WW domains. MAGUKs localizes to regions of cell-cell contact, such as tight junctions in epithelial cells and synaptic junctions in neurons. The MAGI3 gene is associated with the human chromosome at 1p13.2.
Immunogen
C-terminal SLIPR protein fused to GST (amino acids 421-785) of rat SLIPR.
Application
Monoclonal Anti-SLIPR/MAGI-3 antibody produced in mouse may be used in:
- enzyme-linked immunosorbent assay (ELISA)
- immunoblotting
- immunocytochemistry
Biochem/physiol Actions
Membrane-associated guanylate kinase, WW and PDZ domain-containing 3 (MAGI-3) protein serve to position the phosphatase to specific subcellular locations that are involved in the regulation of cell proliferation and survival. This protein interacts with the protein phosphatase and tensin homolog (PTEN) and receptor protein tyrosine phosphatase β (RPTPβ). MAGUKs are involved in the assembly of multiprotein complexes via their protein-protein interaction domains.
Monoclonal Anti-SLIPR/MAGI-3 recognizes rat SLIPR/MAGI-3 (approx. 160 kDa).
Physical form
Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
Preparation Note
For continuous use, store at 2-8 °C for up to one month. For prolonged storage, freeze in working aliquots at −20 °C. Repeated freezing and thawing is not recom-mended. Storage in frost-free freezers is also notrecommended. If slight turbidity occurs upon prolongedstorage, clarify the solution by centrifugation beforeuse. Working dilutions should be discarded if not usedwithin 12 hours.

PTEN/MMAC is a phosphatase that is mutated in multiple human tumors. PTEN/MMAC dephosphorylates 3-phosphorylated phosphatidylinositol phosphates that activate AKT/protein kinase B (PKB) kinase activity. AKT/PKB is implicated in the inhibition of apoptosis, and cell lines and tumors with mutated PTEN/MMAC
