General description

Syntrophins are expressed in a wide range of tissues. α1-Syntrophin (SNTA1) contain two pleckstrin (PH) homology domains, found in a wide array of signaling proteins. SNTA1 gene is located on human chromosome 20q11.2. In rat skeletal muscle, α1-syntrophins are mainly found to be located on the sarcolemma.

Immunogen

synthetic peptide located at the N-terminus of human α1-syntrophin (amino acids 2-18 with C-terminally added lysine) conjugated to KLH.

Application

Anti-α1-Syntrophin (AG-17) antibody produced in rabbit may be used in immunoblotting and immunohistochemistry.

Biochem/physiol Actions

Anti-a1-Syntrophin (AG-17) recognizes a1-syntrophin (60 kDa).
Syntrophins associate with the dystrophin complex to act as modular adapters and helps in recruiting signaling proteins to the membrane. The pleckstrin (PH) homology domains of α1-Syntrophin (SNTA1) mainly helps in mediating this recruiting action. Mutation on this gene is accountable for long QT syndrome 12 (LQT12), an autosomal-dominant manner.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide

Preparation Note

For continuous use, store at 2 °C - 8 °C for up to one month. For extended storage, freeze in working aliquots. Repeated freezing and thawing is not recommended. Storage in "frost-free" freezers is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilution samples should be discarded if not used within 12 hours.
