General description
Granzyme K (GZMK) also known as tryptase 2 (TRYP2), is a trypsin like serine protease, evolutionarily similar to granzyme A (GZMA). In human chromosome, the gene GZMK is localized on 5q11.2.

Biochem/physiol Actions
GZMK is a member of a group of related serine proteases from the cytoplasmic granules of cytotoxic lymphocytes and natural killer cells. GZMK cleave after the basic amino acids like arginine or lysine. Cytolytic T lymphocytes (CTL) and natural killer (NK) cells share the remarkable ability to recognize, bind, and lyse specific target cells. They are thought to protect their host by lysing cells bearing on their surface ′nonself′ antigens, usually peptides or proteins resulting from infection by intracellular pathogens. The protein described here lacks consensus sequences for N-glycosylation present in other granzymes. Gmzk expression level is higher in patients with sepsis and also in bronchoalveolar fluid with respiratory infections. GzmK can cause apoptosis by DNA fragmentation, and also by ROS (reactive oxygen species) production. GzmK triggers cleavage of p53, a proapoptotic protein and that potentiates apoptosis of tumor cells. Gzmk is highly expressed in NK cells and plays a key role in NK cells mediated tumor cell clearance. β-tubulin, a component of microtubule network is cleaved by Gzmk, which is crucial for maintaining the cell shape, mitosis and the intracellular trafficking.
