Gene Information

zebrafish ... spi1b(30117)

General description

PU.1 is a transcription factor that belongs to the erythroblast transformation specific (ETS) family. This protein is highly expressed in hematopoietic cells like mast cells, B lymphocytes, neutrophils, early erythroblasts, and macrophages. PU.1 protein consists of a highly conserved DNA binding domain, a glutamine-rich domain, acidic residues towards the N-terminal, and a central proline-glutamic acid-serine-threonine (PEST) domain.

Immunogen

Recombinant protein encompassing a sequence within the center region of zebrafish PU.1/Spi1.

Application

Suggested starting dilutions are as follows: IHC-Wm: 1:100-1:500. Not yet tested in other applications. Optimal working dilutions should be determined experimentally by the end user.

Biochem/physiol Actions

PU.1 transcription factor facilitates the regulation of myeloid differentiation. This transcription factor is also involved in regulating several other myeloid genes such as macrophage colony-stimulating factor (M-CSF) receptor, CD11 antigen-like family member B (CD11B), granulocyte colony-stimulating factor (G-CSF) receptor, and granulocyte-macrophage colony-stimulating factor (GM-CSF) receptor. Dysregulation of the PU.1 gene results in an increase in erythroblast proliferation, inhibits erythroid differentiation and leads to erythroleukemia.

The transcription factor PU.1 is required for normal blood cell development. PU.1 regulates the expression of a number of crucial myeloid genes, such as the macrophage colony-stimulating factor (M-CSF) receptor, the granulocyte colony-stimulating factor (G-CSF) receptor, and the granulocyte-macrophage colony-stimulating
