General description
Neural precursor cell expressed, developmentally down-regulated 9 (Nedd9), also known as Cas-L and human enhancer of filamentation 1 (HEF1) is an adhesion adapter protein, localized to sites of focal adhesions. It belongs to a larger family of Cas docking adapter proteins including p130Cas and Efs and is expressed preferentially in lymphocytes and epithelial cells. The gene NEDD9 is mapped to human chromosome 6p24.2.
Application
Anti-Nedd9 (C-terminal) antibody produced in rabbit is suitable for immunoblotting and immunofluorescence.
Biochem/physiol Actions
Anti-Nedd9 (C-terminal) specifically recognizes human Nedd9.
Neural precursor cell expressed, developmentally down-regulated 9 (Nedd9) protein participates in integrin and growth factor signaling pathways and regulates growth, motility and apoptosis. It undergoes extensive phosphorylation, a process dependent both on cell adhesion and on the integrity of actin cytoskeleton. In response to integrin receptor binding to the extracellular matrix (ECM), Nedd9 binds to focal adhesion kinase (FAK) and is phosphorylated by FAK and Src kinases. In addition, Nedd9 is regulated at multiple levels in a complex and cell cycle-dependent manner. It is thought to play an important role in early neuronal differentiation and brain development. Nedd9 is also a key player in metastatic cancer. Upregulation of Nedd9 expression is observed in various solid tumors and is greatly implicated in the pathogenesis of BCR-Abl-dependent tumors. Overexpression of Nedd9 has been shown to be required for invasion by glioblastoma and to be strongly linked to the promotion of melanoma metastasis.
Physical form
Solution in 0.01 M posphate buffered saline, pH 7.4, containing 15 mM sodium azide.
Preparation Note
Store at –20 °C. For continuous use, the product may be stored at 2–8 °C for up to one month. For extended storage, freeze in working aliquots at –20 °C. Repeated freezing and thawing, or storage in “frost-free” freezers, is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilutions should be discarded if not used within 12 hours.
