General description

Monoclonal Anti-SORBS3 (mouse IgG2b isotype) is derived from the hybridoma SORB84 produced by the fusion of mouse myeloma cells and splenocytes from BALB/c mice immunized with a synthetic peptide. The gene for sorbin and SH3 domain containing 3 (SORBS3) (also known as vinexin) protein is mapped to human chromosome 8p21.3. This protein belongs to the family of adaptor proteins. All the family members share a sorbin homology (SoHo) domain in the N-terminal region and three Src homology 3 (SH3) domains in the C-terminal region. There are three different isoforms of SORBS3 (α, β, and γ) protein that has been characterized. Isoform α and β contain three SH3 domains while the β isoform does not include the N-terminal SoHo domain. The γ isoform of vinexin has been characterized in mouse fetal gonads. It differs from the α isoform in a few amino acids that are missing from the N-terminal region. Vinexin ⓰ is ubiquitously expressed but vinexin α and γ show tissue-specific expression.

Biochem/physiol Actions

Monoclonal Anti-SORBS3 recognizes human, monkey, and rat SORBS3.
Vinexin α and β participates in cell signaling and the cytoskeletal structure. The α isoform is known to induce actin stress fiber formation and the β isoform plays a role in cell spreading. Vinexin might enhance the activation of c-Jun N-terminal kinase/stress-activated protein kinase (JNK/SAPK) in response to epidermal growth factor (EGF) stimulation by using its third SH3 domain. The growth-factor-induced signal transductions are controlled by the vinexin-son of sevenless (SOS) interactions. The sorbin homology (SoHo) domain has been shown to bind the lipid raft-associated protein flotillin1. The Src homology 3 (SH3) domain allows the binding of specific signaling and cytoskeletal molecules, including vinculin.

Physical form

Solution in 0.01M phosphate buffered saline pH 7.4, containing 15 mM sodium azide.

Preparation Note

Store at –20 °C. For continuous use, the product may be stored at 2–8 °C for up to one month. For extended storage, freeze at –20 °C in working aliquots. Repeated freezing and thawing, or storage in “frost-free” freezers, is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilution samples should be discarded if not used within 12 hours.
