General description
Tensins are a family of focal adhesion proteins that link the extracellular matrix (ECM) and the cytoskeleton via integrins. The tensin family includes four members: tensin 1-4, encoded by different genes. Tensin proteins consists of homologous C1, protein tyrosine phosphatase (PTPase), C2, SH2 and phosphotyrosine-binding domain (PTB) domains. Tensin 2 is very similar to tensin1 at its N- and C-terminus. They have similar actin binding domain, SH2 and PTB domains. Tensin 2 is a binding partner for Axl receptor tyrosine kinase. Tensin 2 mRNA is highly expressed in heart, liver, kidney and skeletal muscle.
Application
Anti-Tensin 2 antibody produced in rabbit has been used in immunoblotting.
Biochem/physiol Actions
Tensins play an important role in regulating cell shape and motility. Abnormal expression of tensins are implicated in various forms of human cancers. Expression of tensin 2 and tensin 3 are markedly reduced in a panel of diverse human cancer cell lines and all four genes are downregulated in human kidney cancer. Tensin 2 induces growth inhibition and apoptosis in hepatocellular carcinoma (HCC) cell lines. Tensin 2 interacts directly with deleted in liver cancer 1 (DLC1), a tumor suppressor protein often deleted and downregulated in HCC cell lines and other types of cancers. Tensin 2 may be important for normal kidney function in vivo.
Physical form
Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
