General description

Monoclonal anti-actin (mouse IgM isotype) is derived from the hybridoma MM2/193 produced by the fusion of mouse myeloma cells and splenocytes from BALB/c mice immunized with human platelet membranes. Monoclonal anti-actin recognizes human, monkey, dog, rat and mouse actin (not tested in other species).
In mammals, actin protein consists of six different isoforms encoded by separate genes. The four isoforms αskeletal-actin, α cardiac-actin, αsmooth-actin and γsmooth-actin are expressed in skeletal, cardiac and smooth muscle. The other two isoforms, β cyto-actin and γ cyto-actin are ubiquitously expressed.

Application

Monoclonal Anti-Actin antibody produced in mouse has been used in immunoblotting.

Biochem/physiol Actions

Actin plays a vital role in various cellular processes, such as cell division, migration, junction formation, chromatin remodeling, transcriptional regulation, vesicle trafficking and cell shape regulation. Polymorphism in the smooth muscle alpha-actin (ACTA2) is associated with the development of thoracic aortic aneurysms and dissections.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide

Preparation Note

Store at -20°C. For continuous use, store at 2-8°C for up to one month. For extended storage, freeze at -20°C in working aliquots. Repeated freezing and thawing, or storage in “frost-free” freezers, is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilution samples should be discarded if not used within 12 hours.

Other Notes

In order to obtain the best results using various techniques and preparations, we recommend determining optimal working dilutions by titration.
