General description
ER degradation-enhancing α-mannosidase like protein 1 (EDEM1) is a putative mannose-binding lectin. EDEM1 is localized to the endoplasmic reticulum (ER) mainly as a soluble glycoprotein.
Immunogen
synthetic peptide corresponding to an internal region within human EDEM1, conjugated to KLH. The corresponding sequence differs by a single amino acid in mouse and by 2 amino acids in rat.
Application
Anti-EDEM1(internal) antibody produced in rabbit has been used in immunoblotting and immunoprecipitation.
Biochem/physiol Actions
Over-expression of ER degradation-enhancing α-mannosidase like protein 1 (EDEM1) leads to endoplasmic reticulum (ER)-associated degradation (ERAD) by stimulating the release of terminally misfolded glycoproteins from calnexin. EDEM degrades misfolded glycoproteins in an N-glycan dependent manner. Three EDEM homologues, EDEM1, EDEM2 and EDEM3 are transcriptionally upregulated by activated inositol-requiring enzyme 1 (IRE1)/X-box binding protein 1 (XBP1) branch. It interacts with the COOH-terminus of calnexin and lacks mannosidase activity.
Physical form
Solution in 0.01 M phosphate buffered saline pH 7.4, containing 15 mM sodium azide.
