General description
Sigma-Aldrich®总黄曲霉毒素 ELISA（酶联免疫吸附测定）试剂盒是一种固相直接竞争性酶免疫测定。一种可与黄曲霉毒素的所有四种亚型发生交叉反应（参见交叉反应信息）的黄曲霉毒素特异性抗体经过优化，包被在聚苯乙烯微孔上。用 70% 甲醇从研磨样品中提取毒素。将提取的样品与 HRP标记黄曲霉毒素B1混合，加入抗体包被的微孔中。提取样品中的黄曲霉毒素与HRP标记黄曲霉毒素B1竞争，与包被在微孔上的抗体结合。倒出微孔内容物，洗涤去除非特异性反应物。加入酶底物 (TMB) 并显色 (蓝色)。颜色深浅与结合标记物的量成正比，与样品或标准品中的黄曲霉毒素浓度成反比。因此，随着样品或标准品中黄曲霉毒素浓度增加，蓝色越来越淡。
