SKY Peptide is a non-cytotoxic and stable nonapeptide modeled after human neutrophil peptide 1 (HNP1, a-defensin) β strand 2 CCL5-binding mode to optimally complete against HNP1 for CCL5 interaction (EC50 ~ 0.1 μg/mL against 10 μg/mL HNP1 and 1 μg/mL CCL5 heteromer formation on HUVECs) without affecting CCL5 and cell surface CCR5 interaction. SKY Peptide is shown to inhibit HNP1-CCL5 heteromer-dependent classical monocytes adhesion both in cultures in vitro (EC50 <1 μg/mL; 10 μg/mL HNP1 and 1 μg/mL CCL5 over HUVEC monolayer) and in mice in vivo.
