General description
Human Pyruvate kinase M2 (PKM2) (GenBank Accession No. NM_002654) full length with N-terminal His tag, MW = 58.9 kDa, expressed in an E. coli expression system.
Application
Useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling.
Biochem/physiol Actions
Pyruvate kinase M2 (PKM2) is an isoform of pyruvate kinase that is expressed in certain differentiated cells and in proliferatively active cells. PKM2 is often overexpressed in cancer cells.
Physical form
Formulated in 25 mM Tris-HCl, pH 8.0, 100 mM NaCl, 0.05% Tween-20, 30% glycerol and 3 mM DTT.
Preparation Note
Thaw on ice. Upon first thaw, briefly spin tube containing enzyme to recover full content of the tube. Aliquot enzyme into single use aliquots. Store remaining undiluted enzyme in aliquots at -70°C. Note: Enzyme is very sensitive to freeze/thaw cycles.
Other Notes
One unit is defined as the amount of enzyme required to transfer a phosphoryl group from phosphoenolpyruvate (PEP) to 1 pmol of ADP/min at 37°C
