Gene Information

human ... GDF9(2661)

General description

GDF-9 is a member of the TGF-β superfamily of growth and differentiation factors, and is highly homologous to GDF-3. Unlike most TGF-family members, GDF-9 and GDF-3 are not disulfide-linked dimers. GDF-3 is expressed in adult bone marrow, spleen, thymus, and adipose tissue. GDF-9 is expressed in oocytes and is required for normal ovarian folliculogenesis. Recombinant human GDF-9 is a 31.0 kDa non-disulfide-linked homodimer containing two 135 amino acid polypeptide chains with 6XHis on its N-terminus.

Application

GDF9 has been as a supplement in the basal medium for the maturation of cumulus–oocyte complexes (COCs).

Biochem/physiol Actions

Growth differentiation factor 9 (GDF9) is known to be localized in the cytoplasm. The gene encoding it is localized on human chromosome 5q31.1. It is known to be associated with granulosa cell proliferation and steroid production. Downregulation of the gene is observed in polycystic ovary syndrome.

Physical form

Sterile filtered and lyophilized with no additives.

Preparation Note

Centrifuge the vial prior to opening. Avoid freeze-thaw cycles.
Reconstitute in water to a concentration of 0.1-1.0 mg/mL. This solution can then be diluted into other aqueous buffers.

Cumulus expansion is essential for ovulation and oocyte maturation in mammals. Previous studies suggest that this process requires certain cumulus expansion enabling factors, induced by LH surge, that activate SMAD signaling locally. However, their identities remain uncertain.
