General description
The PD-10 desalting columns contain Sephadex® G-25 for gel filtration (size exclusion) of biomolecules. Globular compounds pass quickly through the column. Compounds, such as salts, pass through the column slowly. The column actually separates two classes of compounds by their molecular weight, thus desalting the protein of interest.
Application
Disposable PD-10 Desalting Column has been used:
- to concentrate samples of high peroxidase activity from a PGF-HRP (prostaglandin F2α-horse radish peroxidase) conjugate solution and to remove free PGF from this solution.
- for the separation of lanthanide-labeled proteins from unbound lanthanides by column chromatography.
- for repurification of protein by removal of lower molecular weight fragments after elution from Ni-NTA agarose affinity column.
- for separation of Eu-labeled protein from free Eu (Europium).
PD-10 desalting columns have been used to purify reduced Alemtuzumab (Campath, Berlex) humanized IgG1 rat monoclonal antibody from excess phospine, which is being developed for the treatment of chronic lymphocytic leukemia, autoimmune disease and for the prevention of transplant rejection.
