intensity of the color, measured at 610 nm, is directly
proportional to the albumin concentration in the sample.
Components
The kit is sufficient for 250 assays in 96 well plates.
Reagent 50 mL
room temperature, transfer to
cuvette and read OD at 610 nm.
Results
1. Calculate ΔOD by subtracting the reading of
Standard #8 (Blank) from the remaining Standard
reading values.
6. Incubate 10 minutes at room temperature, and
then read the optical density at 595 nm
(550–610 nm).
Procedure Using Cuvette
The following procedure is for use in 1 mL cuvettes;
adjust
clarified lysates on top of the empty manifold. Apply
vacuum at 24 inches of Hg (810 millibar – 610 torr) until wells are empty. Direct filtrate to
waste. When filtration is complete, switch off vacuum
the absorbance of the acceptor solution
at 610 nm. In such instances, transfer 200 µL to a clear bottom plate for analysis. Typical
absorbance values at 610 nm should fall in the range of 0.002 to 0.006
Operating Tips
● Vacuum should remain off while system is being assembled.
● Vacuum pressure of 610–847 millibar (18–25 inches Hg) at 30 L/minute is recommended for optimum performance.
● The waste
(0.02%), NP-40 (0.6%), SDS (0.12%),
Tris (8 mM), and ethanol (0.4%).
Assay Reaction
1. Add 40 L of each standard into wells of a 96 well
plate.
2. Add 40 L
increase with an absorbance
plate reader with path check on at OD of 575 nm (or
ratio of 570 nm/610 nm).
