Detection
Add 0, 25, 50, and 100 L of the 1 mM NH4Cl
Standard Solution into wells of a 96 well plate. Add
ultrapure water to each well to bring the volume to
100 L, generating
10 L
of the Starch Standard (2.0 mg/mL) to 90 L of
ultrapure water, mix well. Prepare Starch Standards in
desired wells of a clear flat-bottom 96 well plate
according to Table 1.

Preparation

Add 0, 30, 60, and 100 µL of the 300 µM Calibrator
solution into tubes. Add water to each tube to bring the
volume up to 100 µL, generating 0 (blank), 90, 180, and
activity.
Standard Curve Preparation
Add 0, 20, 40, 60, 80, and 100 L of 4-Nitrocatechol
Standard into a series of wells in a 96 well plate to
generate 0, 10, 20, 30, 40, and 50 nmole/well of
. Transfer 10 µL Standards into separate wells of a
clear flat-bottom 96-well plate. Add 90 µL Assay
Buffer to the standards.
Detection Reagent Preparation
Prepare enough detection reagents
Solution with 90 L of water to prepare a
10 mM (10 nmole/L) AcAc Standard Solution. Add 0,
2, 4, 6, 8, and 10 L of the 10 mM AcAc Standard
Solution into a 96 well plate to generate 0
