Olive Oil spiked at 25 µg/g with BPA - Weigh 2.0 g of olive oil in a 15 mL screw-cap test tube, add 1 mL of the spike solution (50 µg/mL in, seal with screw cap and shake for 30 seconds. Add 4 mL of diluent and shake for 1 minute. Sonicate the solution for 15 minutes at room temperature and after that centrifuge it at 5000 rev/min for 25 min. Take an aliquot of the supernatant and put it through a 0.45 µm PTFE Filter.

Bisphenol A (BPA) is a precursor to important plastics and can be found in many different consumer goods, e.g. plastic bottles. It is soluble in organic solvents and slightly soluble in water. Due to this characteristic leaching of BPA from plastic bottles into the bottle content can be observed. BPA is a xenoestrogen with the ability to mimic the human hormone estrogen.

This application work is focused on combining HPLC with more sustainable solvents. Therefore, the analysis of BPA content in olive oil was developed by using ethanol as an organic modifier in the mobile phase and as extraction solvent.
For the HPLC separation, a matrix tolerant Chromolith® HighResolution RP-18 endcapped column 100 x 4.6 mm I.D. combined with fluorescence detection (FLD) was applied. A spiked olive oil sample (25 µg/g) was only extracted ethanol and filtered through a 0.45 µm PTFE Filter.
