General description

Adenosine triphosphate (ATP), along with , Guanosine-5′-triphosphate (GTP) , Cytidine triphosphate (CTP) , Uridine triphosphate (UTP), and potentially modified UTP is part of the ribonucleotide mix required for synthesizing messenger RNA (mRNA) during transcription. This process occurs in the presence of RNA polymerase, which catalyzes the formation of RNA from the DNA template using these ribonucleotides.

Application

Guanosine-5′-triphosphate (GTP), cytidine triphosphate (CTP), adenosine triphosphate (ATP), and uridine triphosphate (UTP) are essential nucleotides involved in RNA synthesis. A significant application for these nucleotides is in vitro transcription (IVT) of mRNA, which is widely used in research and experimental studies.IVT kits incorporate these nucleotides to synthesize mRNA from DNA templates, enabling the production of RNA for various applications, including gene expression analysis and functional studies. However, off-the-shelf kits can restrict researchers′ ability to optimize the concentrations and ratios of these nucleotides according to the specific characteristics of different mRNA molecules, such as their composition and length. This limitation can impact the efficiency and quality of the synthesized mRNA. As a result, there is a growing interest in developing customizable approaches that allow for greater flexibility in reagent optimization, enhancing the performance of IVT reactions to better suit specific experimental requirements.
