General description

MDM2单抗（小鼠IgG1同种型）来自小鼠骨髓瘤细胞和BALB/c小鼠脾细胞相互融合而产生的SMP14杂交瘤细胞。鼠类双微体(mdm2)是一种E3泛素连接酶，其N端含有p53结构域。C端结构域则包括真正令人感兴趣的新基因 (RING) 结构域、核定位信号 (NLS)、核仁定位信号 (NoLS) 和核输出信号 (NES)。MDM2定位于人染色体12q15。它表达为不同的可变剪切体。

Biochem/physiol Actions

单独检测 MDM2 蛋白或检测其与 E2F1、p53 和成视网膜细胞瘤蛋白的复合物。与细胞角蛋白 6、14 和 16 观察到轻微的交叉反应。
鼠类双微体 (mdm2) 介导p53抑癌基因的反式激活。MDM2结合组蛋白乙酰转移酶p300转录辅激活因子/组蛋白乙酰酶，从而介导p53降解。MDM2的核定位信号介导其穿过细胞核进入细胞质并介导p53降解。除了与p53的相互作用，MDM2还通过刺激S期诱导转录因子E2F1/二聚化伙伴1 (DP1) 来介导细胞增殖。在胃食管、肝、结肠、乳腺和肺癌中都检测到MDM2基因的扩增。MDM2/p53信号轴的优化状态异常可能会对早衰症产生影响。MDM2特异性单克隆抗体是重要的分析工具，用于确定MDM2的相互作用和分布、信号通路功能以及与p53的相互调节。

Physical form

MDM2单克隆抗体以含15mM叠氮化钠的腹水液形式提供。

Preparation Note

若需连续使用，可在2-8°C下保存最长一个月。若需延长储存时间，可将溶液分装并冷冻。 不建议反复冻融。不建议在无霜冰箱中储存。 如果长期储存时出现轻微浑浊，请在使用前通过离心澄清溶液。 若工作稀释样品在12小时内未使用完，则应丢弃。
