General description

苯甲基磺酰氟(PMSF)是一种广泛使用的丝氨酸蛋白酶抑制剂，可有效抑制胰凝乳蛋白酶、凝血酶和胰蛋白酶等酶。 PMSF通过磺化丝氨酸蛋白酶反应位点的丝氨酸残基中的羟基残基，发挥抑制作用。

Application

苯基甲磺酰氟已用于以下应用：
  • 细胞分级分离。
  • 在核蛋白提取中作为补充剂。
  • 胆固醇酯酶（CE）和假胆碱酯酶（PCE）抑制剂。
  • 在离心之前用于血液的收集以量化血浆ANP的水平。
PMSF常用在裂解缓冲液中，通过抑制蛋白酶、避免其在细胞或组织裂解后降解，从而在蛋白分离和样品制备过程中保留目标蛋白。 PMSF在水溶性培养基中不稳定。 因此，在水溶性培养基中使用前，通常采用无水有机溶剂（例如100％乙醇或无水异丙醇）制备PMSF储备溶液。

PMSF的常见特性和优点包括：
  • 抑制丝氨酸蛋白酶，如胰蛋白酶和胰凝乳蛋白酶

Biochem/physiol Actions
