Gene Information

General description

Placental alkaline phosphatase is found expressed in liver and intestines.

Application

Human placental alkaline phosphatase has been studied extensively due to its ectopic exptression in tumors. It has also been used in a study to investigate whether alkaline phosphatase of human tumor tissue corresponds to one or more phases of trophoblastic development.

Analysis Note

Protein determined by biuret.

Other Notes

One unit will hydrolyze 1.0 μmole of 4-nitrophenyl phosphate per minute at pH 10.4 at 37 ° C.

Alkaline phosphatase electrophoretic patterns characteristic of three phases in early human trophoblast development are described in this preliminary communication. Phase 1 (6 to 10 weeks) consists entirely of two heat-sensitive, L-homoarginine-inhibited bands, the slower one of which possesses antigenic determinants

Alkaline phosphatase (AP) is a widely distributed non-specific phosphomonoesterase that functions through formation of a covalent phosphoseryl intermediate (E-P). The enzyme also catalyzes phosphoryl transfer reaction to various alcohols. Escherichia coli AP is a homodimer with 449 residues per monomer.

Three distinct forms of human alkaline phosphatase, presumably isozymes, are known, each apparently associated with a specific tissue. These are placental, intestinal, and liver (kidney and bone). We have used a specific immunoassay and HPLC to show that placental alkaline
