General description

Monoclonal Anti-MUC1 (mouse IgG1 isotype) is derived from the hybridoma 1D1 produced by the fusion of mouse myeloma cells and splenocytes from BALB/c mice immunized with semi-purified MUC1 from pleural effusion of a breast cancer patient. The Mucin 1 (MUC1) gene encodes a membrane bound glycosylated phosphoprotein, belonging to the family of mucins which are heavily glycosylated proteins. Muc1 is a type I transmembrane glycoprotein with a unique extracellular domain containing 20 amino acid variable number tandem repeat (VNTR) domain. In many epithelia, this protein is attached to the apical surface by a transmembrane domain. The degree of glycosylation varies with cell type. The MUC1 gene is mapped to human chromosome 1q22.

Immunogen

semi purified MUC1 from pleural effusion of a breast cancer patient.

Application

Monoclonal Anti-MUC1 antibody produced in mouse has been used in:
  • immunoblotting
  • enzyme linked immunosorbent assay (ELISA)
  • radioimmunoassay
  • immunoprecipitation
  • immunocytochemistry

Biochem/physiol Actions

Mucin 1 (Muc1) exhibits protective action by binding to pathogens and is also associated with cell signaling capacity. Upregulation of this gene, abnormal intracellular localization and varied glycosylation of Muc1 has been associated with carcinomas. Muc1 might serve as a candidate for single chain antibody mediated targeted therapy in MUC1 expressing cancers. They are preferentially expressed by a variety of adenocarcinomas, including breast, prostate, ovarian and pancreatic carcinomas as well as malignant plasma cells of multiple myelomas.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
