General description

Cytochrome c oxidase subunit 1 (UniProt: P00395;EC:7.1.1.9; also known as Cytochrome c oxidase polypeptide I) is encoded by the MT-CO1 (also known as COI, COXI, MTCO1) gene (Gene ID: 4512) in human. Cytochrome c oxidase subunit 1 (MT-CO1) is a core protein of the cytochrome c oxidase complex, also known as complex IV of the mitochondrial electron transport chain. This protein, encoded by mitochondrial DNA. It comprises of 13 subunits in mammals, with MT-CO1 being one of the three mitochondrial-encoded subunits. MT-CO1 contains multiple transmembrane helices which facilitate its role in electron transport and proton translocation across the mitochondrial inner membrane. MT-CO1 is crucial for the terminal step of the mitochondrial electron transport chain, where it catalyzes the reduction of molecular oxygen to water. In this process, it also assists in maintaining the proton gradient across the mitochondrial membrane by pumping protons from the mitochondrial matrix to the intermembrane space, which is essential for ATP synthesis by ATP synthase. Mutations in the MT-CO1 gene can lead to several mitochondrial disorders, often with a multi-systemic impact due to the crucial role of mitochondria in energy production. Diseases associated with MT-CO1 mutations include Leber′s hereditary optic neuropathy (LHON), which affects vision, and various forms of mitochondrial encephalomyopathy, such as MELAS (Mitochondrial Encephalomyopathy, Lactic Acidosis, and Stroke-like episodes). Application

Quality Control Testing

Evaluated by Western Blotting in HepG2 cell lysate.

Western Blotting Analysis (WB): A 1:1,000 dilution of this antibody detected COXI/MT-CO1 in HepG2 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected COXI/MT-CO1 in mouse heart tissue lysate.

Affinity Binding Assay: A representative lot of this antibody bound COXI/MT-CO1 peptide with at least one thousand-fold (1,000X) higher than with non-specific control peptide.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected COXI/MT-CO1 in human heart tissue sections.

Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected COXI/MT-CO1 in HeLa cells.

Flow Cytometry Analysis: 0.1 µg from a representative lot detected COXI/MT-CO1 in one million SH-SY5Y cells.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Biochem/physiol Actions

Clone 1C22 is a ZooMAb® recombinant rabbit monoclonal antibody that detects COXI/MT-CO1. It targets an epitope within 15 amino acids from the internal region.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Preparation Note

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
