Prostaglandin G/H synthase 1 (UniProt: P23219; also known as EC:1.14.99.1, Cyclooxygenase-1, COX-1, Prostaglandin H2 synthase 1, PGH synthase 1, PGHS-1, PHS 1, Prostaglandin-endoperoxide synthase 1) I encoded by the PTGS1 (also known as COX1) gene (Gene ID: 5742) in human. Cyclooxygenases (COX) are bifunctional hemoproteins that catalyze both the bisoxygenation of arachidonic acid (AA) to form PGG2 and the peroxidative reduction of PGG2 to form PGH2. Hence, COX has two different active sites. On one side, it has the cyclooxygenase active site, and on the opposite side is an entirely separate peroxidase site, which is required to activate the heme groups that participate in the cyclooxygenase reaction. The enzyme complex is a dimer of identical subunits, two cyclooxygenase active sites and two peroxidase active sites. COX-1 is a constitutive enzyme that is associated with the endoplasmic reticulum. It is responsible for maintaining normal physiologic function and is considered as a housekeeping enzyme. Its cyclooxygenase activity oxygenates AA to PGG2, and the peroxidase activity reduces PGG2 to PGH2, the precursor of all 2-series prostaglandins and thromboxanes. This complex transformation is initiated by abstraction of hydrogen at carbon 13 (with S-stereochemistry), followed by insertion of molecular oxygen to form the endoperoxide bridge between carbon 9 and 11 that defines prostaglandins. The insertion of a second molecule of oxygen (bis-oxygenase activity) yields a hydroperoxy group in PGG2 that is then reduced to PGH2 by two electrons. COX-1 is involved in the constitutive production of prostanoids in the stomach and platelets. In gastric epithelial cells, it is a key step in the generation of prostaglandins that play an important role in cytoprotection. In platelets, it is involved in the generation of thromboxane A2, which promotes platelet activation and aggregation, vasoconstriction, and proliferation of vascular smooth muscle cells. Quality Control Testing

Evaluated by Western Blotting in A431 cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected COX-1 in A431 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected COX-1 in lysates from L6 cells an Human small intestine tissue.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected COX-1 in Human small intestine tissue sections.

Affinity Binding Assay:: A representative lot of this antibody bound COX-1 peptide with a KD of 4.4 x 10-8 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected COX-1 in A431 cells.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Clone 1H8-L2 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects COX-1. It targets an epitope within 20 amino acids from the C-terminal region.

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
