General description

NAD(P)H-hydrate epimerase (UniProt: Q8NCW5; also known as EC:5.1.99.6, Apolipoprotein A-I-binding protein (AI-BP), NAD(P)HX epimerase (NAXE), YjeF N-terminal domain-containing protein 1 (YjeF_N1) is encoded by the NAXE (also known as AIBP, APOA1BP, YJEFN1 ) gene (Gene ID: 128240) in human. It belongs to the epimerase family, which catalyzes the epimerization of the S- and R-forms of NAD(P)HX, a damaged form of NAD(P)H, converting it back into its functional form. Structurally, NAXE is a homodimer and it contains domains necessary for binding to NAD(P)H and for its epimerase activity. NAXE is predominantly localized in the mitochondria and is also found in secreted form in extracellular space. It is ubiquitously expressed in various tissues, with higher expression levels observed in metabolically active tissues such as the liver, heart, and kidney. Mutations in the NAXE gene have been linked to a rare autosomal recessive severe neurometabolic disorder characterized by early-onset progressive encephalopathy with brain edema and/or leukoencephalopathy-1 (PEBEL1). These mutations lead to a loss of function of the NAXE protein, resulting in the accumulation of damaged NAD(P)HX, which disrupts cellular redox homeostasis and contributes to the pathogenesis of the disease. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

KLH-conjugated linear peptide corresponding to 17 amino acids from the C-terminal region of human NAXE/AIBP.

Application

Quality Control Testing

Evaluated by Western Blotting in HEK-293T cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected NAXE/AIBP in HEK-293T cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected NAXE/AIBP in lysate from human liver tissue.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected NAXE/AIBP in human kidney tissue sections and human liver tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound NAXE/AIBP peptide with at least ten million-fold (10,000,000X) higher than with non-specific control peptide.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Biochem/physiol Actions

Clone 2E22 is a ZooMAb® recombinant rabbit monoclonal antibody that specifically detects NAXE/AIBP. It targets an epitope within 17 amino acids from the C-terminal region.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Preparation Note

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
