General description

FLAG®-tag, also known as a FLAG octapeptide or FLAG® epitope, is an artificial synthetic peptide tag that is added to proteins using recombinant DNA technology. It is a versatile and widely used tool in molecular biology. The primary function of the FLAG® tag is to facilitate the purification and detection of recombinant proteins. It is recognized by specific anti-FLAG® antibodies, allowing for easy isolation and identification of FLAG®-tagged fusion proteins through various techniques such as Western blotting, immunoprecipitation, ELISA, Flow cytometry and immunofluorescence. The FLAG® -tag is made up of 8 amino acid sequence NH2-DYKDDDDK-COOH (Asp-Tyr-Lys-Asp-Asp-Asp-Asp-Lys). The sequence contains an enterokinase cleavage site. This sequence is derived from the N-terminus of the bacteriophage T7 gene 10 protein. Its small size and minimal impact on protein function make it an ideal choice for a variety of applications, including the study of disease-related proteins. This antibody reacts with N-terminal and C-terminal FLAG® fusion proteins. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Application

Quality Control Testing

Evaluated by Western Blotting with C-terminal FLAG-BAP fusion protein.

Western Blotting Analysis (WB): A 1:10,000 dilution of this antibody detected FLAG® in C-terminal FLAG-BAP fusion protein.

Tested Applications

Flow Cytometry Analysis: 1 µg from a representative lot detected FLAG® tag in one million CHO-S cells transfected with FLAG ®-mTOR protein.

Affinity Binding Assay: A representative lot of this antibody bound FLAG® peptide with at least one thousand-fold (1,000X) higher than with non-specific control peptide.

Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected FLAG® tag in CHO-S cells transfected with FLAG®-mTOR.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Biochem/physiol Actions

Clone 1I8 is a ZooMAb® recombinant rabbit monoclonal antibody that specifically detects FLAG® tagged proteins.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Preparation Note

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
