General description
ZooMAb® antibodies represent an entirely new generation of recombinant monoclonal antibodies. Each ZooMAb® antibody is manufactured using our proprietary recombinant expression system, purified to homogeneity, and precisely dispensed to produce robust and highly reproducible lot-to-lot consistency. Only top-performing clones are released for use by researchers. Each antibody is validated for high specificity and affinity across multiple applications, including its most commonly used application. Major envelope glycoprotein (UniProt: P17501) is encoded by the GP64 (also known as GP67, ORF128) gene (Gene ID: 1403961) in Autographa californica nuclear polyhedrosis virus (AcMNPV). The major envelope glycoprotein GP64 is a class III fusion protein primarily found in baculoviruses, such as the Autographa californica multiple nucleopolyhedrovirus (AcMNPV). GP64 plays a crucial role in viral entry by mediating low-pH-triggered membrane fusion between the viral envelope and host endosomal membranes. Structurally, GP64 undergoes significant conformational changes in response to acidic conditions, transitioning from a pre-fusion state, dominated by β-sheets and a coiled-coil architecture, to a post fusion conformation essential for membrane fusion events. GP64 is indispensable for virion assembly and budding, particularly in the budded virus (BV) phenotype, which facilitates systemic infection within insect hosts. The protein also facilitates efficient intracellular trafficking to the plasma membrane, where it assembles into budding virions. This trafficking involves critical interactions with host proteins such as Rab GTPases (Rab1, Rab4), clathrin heavy chain, and adaptors like AP-1-2β and Snap29, highlighting its integration into vesicular transport pathways. Expression studies reveal that GP64 is localized predominantly at the plasma membrane during viral egress, although its synthesis commences in the endoplasmic reticulum (ER). Notably, the GP64 signal peptide has been shown to activate the ER-associated degradation (ERAD) pathway, which underscores its regulatory role in protein quality control during expression. GP64 has applications beyond baculovirus biology. It is integral to the Baculovirus Expression Vector System (BEVS), a widely utilized platform for recombinant protein production in biomedical research. modified GP64 proteins are being explored for medical applications, such as peptide display for vaccine development. Biochem/physiol Actions
Clone 3I3 is a ZooMAb® rabbit recombinant monoclonal antibody that detects AcV5. It targets an epitope within 9 amino acids from the C-terminal half.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Preparation Note
300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
